One of the people who sent me this game described it as a "real-time 2D worm shooter". This is exactly what Liero is. Two players go head to head using a huge amount of different weapons. There are four different game types: Kill'em All, Game of Tag, Capture the Flag, and Simple CtF. Each of these modes has a wealth of options, invaluable to the game's replayability. There is also an easy to use level editor, but you have to run LEVEDIT.exe to get to it. Instead of attempting to describe all the features and options of Liero, I'll instead suggest you take a look at LIEROENG.txt which is included in the download. It is an excellent manual.

Don't forget to go to the Player 1 and Player 2 options before starting a game. Here you can set the controls and controller (human/computer) of each player, as well as change the name, color, and health. I also recommend speeding up the Cpu Cycles (Ctrl + F12 in DOSBox) to about 5000.
